Jedi-, but not Yoda1-induced activation and diminishes mechanogating of Piezo1. We have
mapped the region that forms the $80 A-long intracellular beam-like structures, which bridge
the blades to the pore. Mutating two single residues in the proximal end of the beam
abolishes Jedi-and Yoda1-induced activation and impairs mechanogating. We propose that
Piezo1 utilizes a lever-like mechanism for effectively transducing force from the
mechanosensing blades to the pore, enabling long-distance gating.
